The Pyrenees Highway is more less an east-west route that links several towns such as Elphinstone, Chewton, Castlemaine, McKenzie Hill, Muckleford South, Newstead, Joyces Creek, Moolort, Carisbrook, Flagstaff, Maryborough, Moonlight Flat, Bung Bong, Avoca, Ampitheatre, Elmhurst, Eversley, Ararat, Dunneworthy, Denicull Creek, Maroona, Willaura, Stavely and Glenthompson.

The route duplexes with A300 (Midland Hwy) at Castlemaine for about 1 km, C288 for about 500 metres at Carisbrook

Eastern Terminus: the junction with the Calder Highway (A79) at Elphinstone.
Western Terminus: the intersection of Glenelg Highway (B160) and Glenthompson-Carramut Road at Glenthompson.
